Inflammatory Myopathies Autoimmune Myositis Polymyositis Dermatomyositis Necrotizing Autoimmune Myopathy Test Indications: Myositis is an inflammatory disease of skeletal muscles, which can be either hereditary or triggered by infections, toxins or immune system disorders. Myositis Profile can be useful for differential evaluation of polymyositis, dermatomyositis, necrotizing autoimmune myopathy, or overlap syndromes associated with connective tissue disease. Laboratory testing for inflammatory myopathies is appropriate in individuals with progressive muscle weakness, particularly in patients with muscular, cutaneous, pulmonary, and articular manifestations, after more common etiologies have been ruled out. |

A negative result does not necessarily exclude autoimmune disease. Positive results should not be interpreted in isolation and must be correlated with relevant clinical findings. Method of Analysis: The Line Blot Immunoassay provides qualitative in vitro determination of human autoantibodies of the immunoglobulin class IgG to the 16 different antigens Mi-2α, Mi-2β, TIF1γ, MDA5, NXP2, SAE1, Ku, PM-Scl100, PM-Scl75, Jo-1, SRP, PL-7, PL-12, EJ, OJ, and Ro-52 in serum. The test kit contains test strips coated with parallel lines of highly purified antigens. In the first reaction step, the immunoblot strips are incubated with diluted patient samples. In the case of positive samples, the specific IgG antibodies (also IgA and IgM) will bind to the corresponding antigenic site. To detect the bound antibodies, a second incubation is carried out using an enzyme-labelled anti-human IgG (enzyme conjugate) catalysing a colour reaction. 1.
